Crithidia Luciliae
The kinetoplast of the haemoflagellate Crithidia luciliae is a rich source of circular dsDNA and provides an excellent substrate when screening for anti-dsDNA autoantibodies.
The presence of these antibodies at high titre is almost exclusively associated with a diagnosis of SLE. The antibody titre correlates well with disease activity in most patients and is a useful marker, both to assess disease severity and to monitor any subsequent response to therapy.
Samples found positive in the Crithidia luciliae assay may be titrated in doubling dilution to obtain an approximation of antibody titre. This kit is recommended as a screening test to identify those patients with autoantibodies to dsDNA.
Clearly, a requirement also exists for a quantitative assay enabling anti-dsDNA titres to be accurately and reproducibly measured.
