Systemic Rheumatic and Connective Tissue Diseases

Rheumatic diseases collectively form a group of disorders that can affect connective tissue, joints, bone or muscle resulting in various diseases including systemic lupus erythematosus (SLE), polymyositis/dermatomyositis, scleroderma, Sjögren's syndrome, rheumatoid arthritis and primary biliary cirrhosis. Distinct profiles of autoantibodies directed to intracellular (cytoplasmic/nuclear) antigens can be detected in these systemic diseases and are included in many sets of diagnostic criteria. BINDAZYME ANA, dsDNA and ENA assays aid in establishing diagnostic and prognostic information for such disorders.

Rheumatoid Arthritis (RA)

Anti-CCP2 autoantibodies appear to be extremely specific for rheumatoid arthritis (RA) (>95%). They occur early in RA and so can be particularly valuable in aiding diagnosis when clinical symptoms may not be easily distinguished from other conditions. An early diagnosis of RA and prompt commencement of appropriate treatment is particularly important if irreparable joint damage is to be prevented. Anti-CCP2 antibodies also show good predictive potential.

Anti-Neutrophil Cytoplasmic Antibodies (ANCA) - Associated Systemic Vasculitis

ANCA are autoantibodies that react with neutrophil specific antigens. The major ANCA antigens in patients with primary systemic small vessel vasculitis are the enzymes myeloperoxidase (MPO) and proteinase-3 (PR3), which are present within neutrophil granules. BINDAZYME EIA MPO and PR3 assays are used to confirm the specificity of ANCA antibodies identified in previous screening tests, e.g. Neutrophil immunofluorescence assays.

Antiphospholipid Syndrome

Antiphospholipid syndrome (APS) is defined by the association between venous and/or arterial thrombosis, recurrent pregnancy loss, thrombocytopenia and the presence of anti phospholipid autoantibodies (aPLs). aPLs are a group of heterogeneous autoantibodies directed against phospholipids such as cardiolipin, phosphatidylserine and co-factor proteins such as beta 2-glycoprotein I (beta 2-GPI). BINDAZYME Phospholipid assays are used to confirm the type and levels of antiphospholipid antibodies to aid in APS diagnosis and treatment.
